砒石对哮喘小鼠血浆8-Isoprostane的影响

摘    要:目的:通过建立小鼠哮喘模型,给予4种剂量砒石治疗,检测血浆8-Isoprostane水平变化,探讨砒石对哮喘小鼠的治疗作用。方法:选用42只昆明种雄性小鼠,随机分为正常对照组和实验组,实验组建立哮喘模型,予地塞米松、砒石治疗,检测肺功能、血浆8-Isoprostane水平及BALF中WBC计数。结果:砒石治疗组小鼠t/tPmax降低(P<0.05),哮喘组BALF中白细胞总数较正常对照组显著升高(P<0.001);经地塞米松和不同剂量砒石治疗后白细胞总数显著下降(P<0.001);哮喘组血浆中8-Isoprostane较正常对照组显著升高(P<0.001),经地塞米松和不同剂量砒石治疗后均显著下降;8-Isoprostane下降与白细胞计数呈正相关性。结论:哮喘小鼠机体存在氧化应激状态,砒石能够减轻其气道阻塞,降低气道高反应性,纠正氧化/抗氧化失衡。

Abstract:Objective: Through the establishment of mouse' ovalbumin-sensitized asthmatic model and the observation of the 8-Isoprostane of plasm,to evaluate the therapeutic effects of arsenolite on asthmatic mice. Method: Forty-two healthy Kunming male mice were randomly divided into control group and experience groups,the latter were treated with dexamethasone,arsenolite.Lung function were tested,8-isoprostane of plasm and WBC of BALF were measured.Result: Lung function improved after treating with dexamethasone or arsenolite.The WBC of asthmatic mice were significantly higher than those in control group,and decreased after treating with dexamethasone or arsenolite;8-Isoprostane of plasm in asthmatic mice was higher than that of control group,and decreased after treating with dexamethasone or arsenolite.Conclusion: There is oxidant stress status in asthmatic mice.Arsenolite could lighten airway obstruction,reduce airway high response and redress oxidant stress status in asthmatic mice.
