多发性硬化患者外周血T淋巴细胞凋亡和Fas的表达

摘    要:目的 探讨多发性硬化(MS)患者外周血T淋巴细胞凋亡和Fas表达及其与复发缓解病程的关系。方法 42例复发缓解型MS患者,根据其病程分为复发期组20例和缓解期组22例,另外选择20例健康人作为对照组,采用五色流式细胞术(FCM)检测以上三组外周血T淋巴细胞凋亡和Fas表达的情况。结果三组间外周血T淋巴细胞Fas的表达无明显差别。复发期MS患者凋亡的CD4^+T淋巴细胞(由AV染色标记)和Fas相关的CD4^+T淋巴细胞凋亡(由AV^+CD95^+/CD4^+标记)明显低于缓解期MS患者和对照组(均P〈0.05)。复发期MS患者和缓解期MS患者凋亡的CD8^+T淋巴细胞(由AV染色标记)和Fas相关的CD8^+T淋巴细胞凋亡(由AV^+CD95^+/CD8^+标记)都明显低于对照组(均P〈0.05),但复发期MS患者和缓解期MS患者之间无明显差异(P〉0.05)。结论T 淋巴细胞的凋亡和Fas的表达与复发缓解型MS患者的发病和病情的加重有关,MS患者对Fas/FasL介导凋亡的抵抗可能参与了MS发病的调节机制。

Abstract:Objective To explore the relationship among Fas(CD95) expression in T lymphocyte,Fas-mediated apoptosis and the disease course of multiple sclerosis(MS).Methods Forty-two patients with relapsing-remitting MS were divided into relapsing MS group(n=20) and remitting MS group(n=22),and 20 healthy subjects were selected as controls.Annexin V(AV) binding and Fas expression in T cells in peripheral blood from MS patients and controls were examined in all groups by five-color flow cytometry.Results Fas expression in T lymphocyte had no significant difference between 3 groups.The levels of AV CD4 CD3 cells(labeled by AV) and CD95 AV CD4 CD3 cells(labeled by CD95 and AV) in peripheral blood were significantly lower in relapsing MS group than in remitting MS group and control group.The levels of AV CD8 CD3 cells(labeled by AV) and CD95 AV CD8 CD3 cells(labeled by CD95 and AV) in peripheral blood were significantly lower in relapsing MS group and in remitting MS group than in control group,but there was no significant difference between relapsing MS group and remitting MS group.Conclusion These results indicate that the resistance of CD4 T cells to Fas-mediated apoptosis is involved in exacerbation of MS,that Fas-mediated apoptosis of T cells is associated with remission of MS,and that the resistance of T cells to Fas/FasL-mediated apoptosis is involved in regulatory mechanisms of MS.
