山奈酚对糖尿病性微血管内皮细胞RAGE表达的影响

摘    要:目的:观察山奈酚(kaempferol)对糖尿病性心肌病微血管内皮细胞AGE受体(RAGE)表达的作用。方法:体外原代培养心脏微血管内皮细胞,采用牛血清白蛋白糖基化终末产物(BSA-AGEs)(100mg/L)与山奈酚(2μM,10μM,50μM)作用于心脏微血管内皮细胞(24h),免疫组化法和western法检测RAGE蛋白表达。结果:随着山奈酚浓度的增加,RAGE蛋白表达逐渐减少(P<0.05)。结论:山奈酚能够抑制AGEs诱发的心脏微血管内皮细胞RAGE的表达,从而进一步抑制氧化应激的发生。

EFFECTS OF KAEMPFEROL ON AGES-INDUCED RAGE IN CARDIAC MICROVASCULAR ENDOTHELIAL CELLS

Abstract:Objective: To observe the effects of kaempferol on AGEs - induced RAGE production in cardiac microvascular endotheli- al cells. Methods:to culture the cardiac microvascular endothelial cells in vitro, respectively, AGEs (100mg/l) was added and re- sumed incubation for 24 hrs. Meanwhile, a group managed with serum - free culture solution was assumed as blank control. We deter- mined the RAGE protein expression of all groups. Results:As kaempferol concentration was increased, RAGEs protein expression was seen decreased( P 〈 0. 05). Conclusion: Kaempferol may inhibit intracellular RAGEs protein expression in cardiac microvascular endo- thelial cells, furthermore to suppress activation of oxidative stress pathway.
