Tissue distribution of hydrazine in mice
Abstract:Mice which had been administered hydrazine(Hz) 14 mg·kg -1 iv were killed at 5, 15, 30, 60, 120 and 240 minutes and blood, heart, liver, spleen, lungs, kidneys, brain, muscles and intestine samples were collected. The tissues were homogenized in 4 volumes of 0.1 mol·L -1 HCl. The concentration of Hz in blood and the homogenate of tissues was assayed by a p-dimethylaminobenzaldehyde (p-DMBA) colorimetric method. The distribution characteristics of Hz in mice were quantitatively compared by means of the parameters of targeting efficiency (t e, t e=(AUC ∞ 0) Tissue (AUC ∞ 0) Blood ) and weighted-average targeting efficiency (t * e, t * e=(AUQ ∞ 0) Tissue (AUQ ∞ 0) Blood ) The concentration-time data for Hz in various tissues of mice could be fitted by the exponential relationship or biexponential relationship. Total areas under Hz concentration-time curves(AUC ∞ 0) of blood, heart, liver, spleen, lungs, kidneys, brain, muscles and intestine were 6.3, 6.5, 13.2, 10.7, 8.7, 43.9, 3.4, 6.1 and 6.9 μg·h·g -1 , respectively; total areas under amount-time curves(AUQ ∞ 0) of these tissues(not including muscles)were 9.7, 0.6, 11.8, 0.8, 1.6, 10.2, 1.4 and 18.3 μg·h·g -1 , respectively. The t * e values for liver and kidneys were 2.1 and 7.0, respectively,indicating that Hz was selectively distributed to liver and kidneys; however, the t * e values of tissues indicated that intestine and liver contained higher amount the tissues of Hz.
