摘    要:目的探讨高良姜高效液相色谱法指纹图谱(HPLC—FPs)分离的优化条件。方法色谱柱为Shim-pack VP-ODS C18柱(150mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸水溶液,检测波长为210nm,柱温为30℃,考察高良姜体积分数为90%的乙醇提取物在不同梯度洗脱条件下的分离情况。结果优选出的高良姜HPLC—FPs分离条件,可使高良姜体积分数为90%的乙醇提取物中各组分达到较好分离,稳定性、精密度、重现性好。结论该色谱分离条件可用于高良姜药材HPLC—FPs的分离。

Abstract:Objective To optimize the separation conditions of fingerprint for Alpinia officinarum Hance by HPLC. Methods RP- HPLC method was adopted. The samples were extracted by 90% ethanol (V/V) solution and separated by the linear gradient elution on Shimpack VP- ODS C 18 column( 150 mm× 4. 6 mm, 5 μm), with acetonitrile- 0. 1% phosphoric acid solution as the mobile phase. The eluent was detected at UV 210 nm and the column was maintained at temperature 30℃. During the linear gradient elution program, the volume fraction of acetonitrile in the mobile phase changed as follows: 0-60 min, from 11% to 50% ; 60-90 min, from 50% to 100% ; 90- 100 min, 100%;the flow rate was 1 mL/min. Results The separation conditions described above achieved good result in the aspects of separation effect, stability, sensitivity, and reproducibility. Conclusion The optimum separation conditions of fingerprint would be useful for establishing the method of fingerprint for Alpinia officinarum Hance by HPLC.
