摘    要:目的研究新化合物O325H对2型糖尿病的作用。方法灌胃给药,观察O325H对正常小鼠的糖耐量以及对两种2型糖尿病小鼠的降糖降脂作用。采用RT PCR方法,研究O325H的作用机制。结果O325H对正常小鼠的糖耐量没有明显影响,对2型糖尿病小鼠有明显的降糖降脂作用,同时能够增加不同组织中脂蛋白脂酶(LPL)、脂肪酸转运蛋白(aP2)和葡萄糖转运体4(GluT4)mRNA的表达。结论O325H具有良好的降糖降脂作用,其作用机制符合PPARα/γ双重激动剂的特点,具有成为预防和治疗2型糖尿病药物的潜力。

Abstract:Objective To study the effect of O325H on type 2 diabetes.Methods The effects of O325H were investigated in normal KM mice and two kinds of type 2 diabetic mice by intragastric gavage.The possible mechanisms of O325H were investigated by using RT-PCR.Results O325H couldn′t decrease blood glucose in normal mice.However,it could reduce blood lipid level and improve glucose metabolism in type 2 diabetic mice.O325H also increased the expressions of LPL,aP2 and GluT4 mRNA in several tissues.Conclusions O325H is a novel PPARα/γ agonist,which have a great effect to reduce blood lipid and glucose.These results show that O325H has great potential as a prevention and therapy for type 2 diabetes..
