以Ecarin测定血浆中凝血酶原的活性

摘    要:目的 :建立一种准确、快速和灵敏度较高的检测血浆中凝血酶原活性的方法。方法 :以巨齿蛇毒中的有效成分Ecarin作为凝血酶原的激活剂 ,分别采用Ecarin凝固时间法 (ECT)和发色底物法测定血浆中的凝血酶原的活性。结果 :Ecarin浓度在 1~ 8u ml范围内 ,随着血浆的稀释 ,ECT逐渐延长 ,ECT(lg)和血浆稀释度 (lg)之间的相关系数均在 - 0 .94 3以上 ,P <0 .0 1;Ecarin浓度 (lg)和ECT(lg)的相关系数均在 - 0 .95 7以上 ,P <0 .0 1。血浆稀释度为 1:5 0、1:10 0、1:2 0 0、1:4 0 0时 ,Ecarin发色底物法所测血浆稀释度 (lg)与A值 (lg)之间的相关系数γ =- 0 .974(P <0 .0 1)。结论 :Ecarin凝固时间法和发色底物法均具有良好的灵敏度 ,能较好地反映血浆中凝血酶原活性的变化

Abstract:Objective To develop a highly accurate and sensitive test for the determination of the activity of prothrombin in human plasma. Methods Ecarin, a protease extracted from Echis carinatus venom ,was used as the activator of prothrombin, Ecarin clotting times(ECT) and chromogenic substrate method were used. Results When the concentration of Ecarin was in the range of 1~8 u/ml, ECT gradually prolonged with the dilution of the plasma. The correlation(γ) between ECT (lg) and plasma dilution(lg) was more than -0.943, P<0.01;the correlation(γ) between the concentration of Ecarin(lg) and ECT(lg) was more than -0.957, P<0.01. When the dilution of the plasma was 1:50,1:100,1:200,1:400,the correlation(γ) between plasma dilution(lg) and value A(lg) was -0.957, P<0.01. Conclusion Both the ECT and chromogenic substrate method were sensitive to determine the activity of prothrombin in plasma.
