白藜芦醇对衰老昆明小鼠肝细胞作用机制的探讨

摘    要:目的:观察白藜芦醇对衰老雌性昆明小鼠肝细胞凋亡相关蛋白Bcl-2、Bax及其抗氧化能力的影响,研究白藜芦醇延缓肝细胞衰老的作用和机制。方法:采用自然衰老雌性昆明小鼠,给予不同剂量的白藜芦醇,用免疫组化方法观察肝细胞凋亡相关蛋白Bcl-2、Bax的表达,用生化法测定血清总超氧化物歧化酶(SOD)活力和丙二醛(MDA)含量。结果:衰老对照组与青年对照组比较,Bcl-2表达明显降低,Bax表达升高,血清中SOD活力下降、MDA含量显著升高(P〈0.05),120mg/ml白藜芦醇可以使Bcl-2表达及SOD活力升高,Bax表达降低,MDA含量显著减少(P〈0.05)。结论:120mg/ml白藜芦醇可调控凋亡相关蛋白Bcl-2、Bax的表达和对抗自由基对肝细胞的损伤,对肝细胞具有一定的保护作用。

Abstract:Objective:To study the effect of resveratrol on hepatocellular apoptosis-related protein Bcl-2 and Bax of aging Kunming mice and its impact on antioxidant capacity in female mice,and study the senility-delaying effect and mechanism of resveratrol on hepatic cells. Methods:Natural aging female mice were given different doses of exogenous resveratrol. Then apoptosis regulatory protein Bcl-2 and Bax expression in mice hepatocellular grandlose cells were detected by using immunohistochemical method. The contents of serum total superoxide dismutase activity and malondialdehyde were detected by using biochemical method. Results:Compared with the young control group,the resveratrol of Bcl-2 expression in aged control group was lower and the resveratrol of Bax expression was higher,serum SOD activity was decreased and the resveratrol of MDA was significantly increased (P〈0.05). The Bcl-2 expression and the activity of SOD were also increased in 120 mg/ml resveratrol group,the Bax expression and MDA were significantly decreased (P〈0.05). Conclusion:The 120 mg/ml resveratrol can regulate the expressions of apoptosis-related protein Bcl-2 and Bax,and confront free radical damage which contributes to a protective effect on hepatic cells.
