反相高效液相色谱法定量分析旋覆花中1,5-二-O-咖啡酰奎尼酸

摘    要:目的建立旋覆花药材中1,5-二-O-咖啡酰奎尼酸含量的HPLC测定方法。方法色谱柱为AgilentEclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm);流动相甲醇-0.1%冰醋酸水溶液〔32∶68(0 min)~32∶68(15 min)~36∶64(40 min)〕梯度洗脱;检测波长320 nm;柱温30℃;流速1.0 ml/min。结果 1,5-二-O-咖啡酰奎尼酸在0.62~12.48μg(r=0.999 9)具有良好的线性;平均加样回收率(n=6)为100.9%(RSD=1.13%),方法精密度、稳定性、重复性好。10批药材中1,5-二-O-双咖啡酰奎尼酸的含量范围为1.03%~2.09%,平均含量为1.53%。结论该定量分析方法可用于旋覆花药材的质量控制。

Abstract:Objective A method was established for quantitative analysis of 1,5-di-O-caffeoylquinic acid in Inula japonica Thunb.by RP-HPLC.Methods The analysis was performed on an Agilent Eclipse XDB-C18 analytical column(4.6 mm × 250 mm,5 μm) with the mobile phase of methanol and water(0.1 % CH3COOH)in gradient mode(32 ∶ 68(0 min)-32 ∶ 68(15 min)36 ∶ 64(40 min)).And the flow rate was 1.0 ml/min with the detection wavelength of 320 nm as well as the column temperature at 30 ℃.Results The linear equation was determined as Y=2581.50 X-326.15(r= 0.999 9).And the average recovery and RSD were 100.9 % and 1.13 %.The precision,reproducibility and stability were all consistent with the request of quantitative analysis.The content range of 1,5-di-O-caffeoylquinic acid in 10 samples was determined as 1.03 %-2.09 %,average content was 1.53 %.Conclusion The established method is good for the quality control of Inula japonica Thunb.
