FasL基因治疗移植于裸鼠的人脑胶质母细胞瘤的实验研究

摘    要:目的 :探讨FasL基因在体内诱导胶质瘤细胞凋亡和抑制增殖的作用。方法 :在裸鼠皮下种植Fas表达阳性的人脑胶质母细胞瘤细胞系TJ90 5 ,成瘤后 ,利用脂质体介导FasL基因瘤内治疗 ,在 3 9d观察期间 ,用千分尺测量肿瘤生长 ,最终取出肿瘤 ,用原位杂交 ,免疫组化鉴定FasL基因和蛋白的表达 ,应用TUNEL法检测细胞凋亡 ,Ki 67免疫组化检测细胞增殖。结果 :FasL基因瘤内注射观察 3 9d后 ,治疗组瘤体积明显小于对照组 (P <0 0 1) ,肿瘤组织细胞内FasL基因表达增加 ,凋亡细胞数增多 ,细胞增殖率降低。结论 :FasL基因对Fas表达阳性胶质瘤有治疗作用。

Abstract:Aim:To study the apoptosis-inducing and proliferating-inhibitory effect of FasL gene on human glioma cells in nude mice.Methods:TJ905 human glioblastoma cells with positive Fas expression were implanted into nude mice subcutaneously,when the tumors were established,FasL gene was injected intratumorally mediated by lipofectamine.During observation of 39 days,the tumor growth was measured consecutively and the tumor mass was taken out at the 39 day ,the FasL gene and protein expression were examined by using in situ hybridization and immunohistochemical staining.The cell apoptosis and the proliferating activity were examined by TUNEL and Ki-67 immunohistochemical staining method,respectively.Results:After 39 days,the tumor volume of treated group was smaller than that of the control and empty plasmid treated group (P<0.01).FasL gene expression,the number of tumor cells with apoptosis and the effect of proliferating inhibition in treated group were more significant than that of the other two groups.Conclusion:FasL gene therapy has some inhibitory effects on the growth of Fas positive human gliomas.
