摘    要:本文报道用离子对反相液相色谱—电化学检测法同时测定大鼠脑内单胺递质及其代谢产物。本法采用高氯酸沉淀组织蛋白后上清液直接进佯技术,能使NE、E、DA、5-HT及它们的主要代谢产物MHPG、DHPG、HVA、DOPAC、5-HIAA与内标异丙肾上腺奈等十种物质很好地同时分离。由此不仅能测定单胺类神经递质在脑内的含量,并能测定其转换率。方法简便快速,回收率较高且稳定。文中对色谱分离及检测的某些影响因素做了分析讨论,并测定了大鼠下丘脑内单胺递质及其代谢产物。

Abstract:This paper reports a reversed-phase high-performance liquid chromatography with electrochemical detection to separate and determine monoamines and their metabolites (norepinephrine, epinephrine, dopamine, 5-hydroxytryptamine, 3,4-dihydroxyphenylglycol, 3-methoxy-4-hydroxyphenylglycol, 3,4-dihydroxyphenyacetic acid, homovanillic acid and 5-hydroxyindole-3-acetic acid) simultaneouslyl in rat brain, using sodium octyl sulfate and dibutylamine as counter-ions, and isoproterenol as internal standard. The supernatants of protein-precipitated rat brain tissue homogenates were directly injected into the chromatographic system. The chromatographic conditions such as mobile phase pH, concentration of methanol, counter-ions, and applied potential of working electrode, etc., were discussed. The monoamines and their metabolites in rat hypothalamus were determined.
