端粒酶蛋白hTRT基因表达在小肝癌诊断中的价值

摘    要:目的 :探讨端粒酶蛋白hTRT基因表达在小肝癌诊断中的价值。方法 :选择具有完整病理资料的 2 0例小肝癌 ,用原位杂交、免疫组化技术检测端粒酶蛋白hTRT基因、P5 3蛋白和PCNA的表达 ,然后将hTRT的表达与小肝癌病理表现对照。结果 :2 0例小肝癌中hTRT阳性率为 80 % (16 / 2 0 )、P5 3阳性率为 35 % (7/ 2 0 )、PCNA强阳性率为 5 5 % (11/ 2 0 ) ,3种指标在癌中表达强度显著高于癌旁组织中表达 (P <0 .0 5 ) ,癌旁相对正常组织中未检出阳性。hTRT阳性率与病理表现无显著关系 (P<0 .0 5 )。结论 :在小肝癌发生发展的多基因作用中 ,端粒酶对肝细胞的增生和恶变起重要作用 ,端粒酶蛋白hTRT基因检测有助于小肝癌的早期诊断

Abstract:Objective: To study the value of telomerase activity in the diagnosis of small hepalocellular carcinoma (SHCC). Methods: 20 small HCC and adjacent liver tissue were examined for telomerase protein hTRT gene, P53 and PCNA by in situ bydridization(ISH) tecnigue ,and comparison of its pathology. Results: And P53 were respectively 80%(16 /20) and 35%(7 /20), the strong positive rate of PCNA was 55%(11 /20).The position level were siginificsnt higher than its adjacent tissue(P<0.05),while negative response appearing in relative normal liver.No significant relationship could be observed between the positivity of telomerase and fingings of pathology(P<0.05). Conclusion: These results indicate that activation of telomerase may be required as a critical step in hepatocarcinogenesis and tumor development,and detection of telomerase activity may be useful in diagnosis of SHCC.
