2型糖尿病微血管病变患者血浆对氧磷酯酶-1活性与氧化应激的关系

摘    要:目的探讨2型糖尿病微血管病变(DMPA)患者血浆对氧磷酯酶-1(PON-1)活性与氧化应激的关系。方法168例2型糖尿病根据微血管并发症情况分为无微血管并发症(NDC)组、糖尿病视网膜病变(DR)组和糖尿病肾病(DN)组,分别检测各组患者血浆PON-1、超氧化物歧化酶(SOD)和循环谷胱甘肽过氧化物酶(GSHPx)活性、丙二醛(MDA)含量,分析PON-1活性与SOD、MDA、GSHPx之间的关系。结果糖尿病各组患者血浆PON-1、SOD、GSHPx活性均明显低于对照组(P<0.01),DMPA组又低于NDC组(P<0.05),而血浆MDA含量与对照组比较显著升高(P<0.01),DMPA组又高于NDC组(P<0.05)。相关分析表明,糖尿病患者血浆PON-1活性与GSHPx、SOD呈正相关(r=0.774,P<0.01;r=0.801,P<0.01),与MDA呈负相关(r=-0.833,P<0.01)。结论在2型糖尿病DMPA中氧化应激增强及血浆PON-1活性显著降低,PON-1与氧化应激及其相互作用参与了2型糖尿病DM-PA的发生和发展。

Abstract:Objective To investigate the relationship between the avtivity of plasma paraoxonase-1(PON-1)and oxidative stress in type 2 diabetes with microangiopathy.Methods 168 cases of type 2 diabetes mellitus were divided into three groups according to vessel lesions:no-diabetic complication group(NDC),diabetic retinopathy group(DR)and diabetic nephropathy group(DN),and matched with normal controls(NC).The plasma activities of PON-1,superoxide dismutase(SOD),glutathione peroxidase(GSHPx),and the contents of malondialdehyde(MDA)were measured in the type 2 diabetes mellitus and 48 normal controls.The analysis of linear correlation was performed to estimate the relationship between these variables.Results The plasma activities of PON-1,GSHPx,SOD were significantly lower in patients with type 2 diabetes mellitus than in the control subjects(P<0.01).The plasma MDA levels were significantly higher in patients with type 2 diabetes mellitus than in the control subjects(P<0.01).The plasma activities of PON-1,GSHPx,SOD were significantly lower in DMAP than those without microangiopathy(P<0.05).The plasma levels of MDA were significantly higher in DMAP than those without microangiopathy(P<0.05).The analysis of linear correlation showed PON-1 was positively correlated with GSHPx(r=0.774,P<0.01),SOD(r=0.801,P<0.01)and negatively correlated with MAD(r=-0.833,P<0.01).Conclusion Plasma PON-1 activities were significantly decreased in type 2 diabetes with microangiopathy.Oxidative stress may participate in deterninining PON-1 activity in type 2 diabetes with microangiopathy.The conmlex interaction between oxidative stress and PON-1 may result in accelerated microangiopathy in type 2 diabetes mellitus.
