原位杂交检测增生和癌变前列腺组织中前列腺分泌蛋白94 mRNA的表达

摘    要:根据前列腺分泌蛋白 94(PSP94)和PSP5 7的基因结构特点 ,设计并合成了以地高辛标记的两条探针 ,利用组织原位杂交的方法 ,对增生和癌变前列腺组织中PSP94mRNA的表达量进行了比较。结果显示 ,2 0例增生前列腺组织 ,有 1 0例PSP94和PSP5 7共同探针的杂交信号比PSP94特异探针杂交信号强 ,1 0例相反。它们的共同点是均以腺体增生为主。而 7例前列腺癌组织中除 1例外 ,其余 6例PSP94特异探针的杂交信号均比PSP94和PSP5 7共同探针的杂交信号强。增生和癌变前列腺组织杂交结果比较 ,癌变前列腺组织PSP94特异探针杂交信号均比增生前列腺组织强。

Abstract:In order to analyse the sense of diagnosis of the PSP94 in prostate carcinoma and benign prostatic hyperplasia (BPH), we designed and synthesized two probes marked by DIG according to the features of the structures of the genes of PSP94 and PSP57. We compared the expression of the PSP94 mRNA in prostate carcinoma and benign prostatic hyperplasia by in situ hybridization. In benign prostatic hyperplasia the result showed the signal produced by the PSP94 and PSP57 probe was stronger than the special PSP94 probe in 10 cases, and was weaker in the other 10 cases. Most were gland hyperplasia. In prostate carcinoma that the signal produced by the special PSP94 probe was stronger than in six cases except one the other probe. The hybridization signal in the prostate carcinoma is stronger than in the BPH.
