瓣膜支架上种植内皮细胞的离体研究

摘    要:目的研究人工培养的内皮细胞在瓣膜支架上的生长情况及黏附力。方法脐带静脉内皮细胞在体外培养扩增,将培养的内皮细胞种植在经激光处理的瓣膜支架上,培养2,4,6,8d后扫描电镜观察内皮细胞在瓣膜支架上的生长情况;并进行剪切力试验,观察内皮细胞的黏附力。结果培养8d后,内皮细胞可在瓣膜支架上形成细胞单层,经模拟血流冲击后,仍有部分内皮细胞粘附在瓣膜支架表面。结论体外扩增内皮细胞种植瓣膜支架是一种可行的方法。

Abstract:Objective To study growth and adherent properties of endothelial cells seeded in artifical valve. Methods Isolating the endothelial cells from human umbilical vein, then cultured and passaged. The endothelial cells were seeded on the surface of valves for co-culture with endothelial cells. The valves sections were examined with scanning electron microscope after 2,4,6,8 days. The valve were washed with pulsatile flow of culture medium before examination. Results After 2,4 and 6 days cultured, the valves were covered by endothelial cells in 20%-30%, 50%-60% and 70%-80% respectively. After 8 days, a monolayer of endothelium was formed on the surface of the valves. There were still a lot of endothelial cells on the valve surface after being washed. Conclusion Endothelialization of the valves is worthy for further clinical trial.
