坐骨神经周围注射可乐定对CCI大鼠痛域的影响

摘    要:目的 观察损伤坐骨神经周围注射可乐定对慢性坐骨神经结扎模型(chronic constriction injury ,CCI)大鼠痛域影响,探讨α2-肾上腺素受体激动剂是否对已经形成的神经病理性疼痛疼痛产生作用。方法 雄性SD大鼠32只,随机分为4组,每组8只。分别为:Sham组:只分离皮肤、肌肉,不结扎坐骨神经; Sal组:形成CCI后注射生理盐水组; Clo组:形成CCI后注射可乐定组; Clo+Yo组:形成CCI后注射可乐定和育亨宾组。采用热辐射法和von Frey细丝法分别测定大鼠热刺激缩足反射潜伏期(thermal withdrawal latency ,TWL)和机械刺激缩足反射痛阈值(mechanical withdrawal threshold ,MWT)。结果 CCI大鼠在术后从7 d开始表现出明显的热痛敏和机械痛敏,损伤坐骨神经周围注射可乐定能明显减轻热痛敏和机械痛敏(р<0.01)。结论 外周神经周围注射α2-肾上腺素受体激动剂可以减轻已经形成的神经病理性疼痛痛域,α2-肾上腺素受体可能参与神经病理性疼痛的发展和转归。【关键词】 可乐定 外周注射 CCI大鼠 神经病理性疼痛 热痛敏 机械痛敏

The effect of administered Perineural clonidine on the pain thresholdof the rats following chronic constriction injury(CCI) of the sciatic nerve
Abstract:【Abstract 】Objictive To investigate the role of perineural clonidine on the pain threthold and the expression of TNF-α and IL-6 of the rats following chronic constriction injury(CCI) of the sciatic nerve .MathodsThirty-two male SD rats were randomly divided into fourgroup:shamgroup ;Sal group :CCI+ saline; Clo group: CCI+clonidine; Clo+Yo group :CCI+clonidine+ yohimbine,n=8.Rats underwent CCI, and perineural saline, clonidine, yohimbine or yohimbine plus clonidine were injected 7 days later . we measured the mechanical withdrawal threshold (MWT) and thermal withdrawal latency(TWL)of the rats after injection . Results The rats of CCI group demonstrated steady mechanical and heat hyperaglsia from the seventh postoperative day.Perineural clonidine reduced MWT and MWT in CCI rats(р<0.01).Conclusion administered Perineural clonidine on the pain threshold of the rats following chronic constriction injury(CCI) of the sciatic,α2-adrenoceptor agonist participate to the development and turnover of neuropathic pain.
