Abstract:Objective To establish an high performance liquid chromatography method for the determination of vancomycin and norvancomycin in human plasma.Methods Vancomycin and norvancomycin were used as the mutual internal standard.The content of mycophenolic acid was detected by ultraviolet detector at 236 nm with Diamonsil C18(2)(150 mm×4.6 mm,5 μm)column at a flow rate of 1.5 ml/min.The mobile phase was 0.05 mol/L NaH2PO4(pH=3.2)-methol(87∶13).The column temperature was 35℃.Results The linear range was 5-200 μg/ml.The within-day RSD was less than 5%,and between-day RSD was less than 7%.The absolute recovery was higher than 60%,and the relative recovery was higher than 90%.Conclusion This method is simple,sensitive,reproducible,and suitable for the therapeutic drug monitoring and the pharmacokinetic study of vancomycin and norvancomycin.
