Abstract:Objective To evaluate the potential actions of angiotensin converting enzyme (ACE) inhibitors on blood vessels.Methods By means of ex vivo mesenteric artery perfusion, HPLC and RIA, aldosterone in the perfusate was measured in normal ( n =10) and perindopril (a tissue penetrative ACE inhibitor) treated ( n =10) groups. The expression of aldosterone synthase gene CYP11B2 mRNA in the vessels was determined by RT PCR and Southern blot.Results Perindopril inhibited aldosterone production and CYP11B2 mRNA expression in the blood vessels. Conclusion The inhibition of aldosterone biosynthesis is involved in the remedy of hypertensive vascular damage by perindopril.
