丹红注射液对大鼠心肌缺血再灌注损伤的保护作用

摘    要:目的观察大鼠心肌缺血再灌注损伤模型心肌组织中超氧化物歧化酶、谷胱甘肽过氧化物酶、诱导型一氧化氮合酶、总一氧化氮合酶活性及一氧化氮、丙二醛含量的变化,探讨丹红注射液对离体大鼠缺血再灌注心肌的保护作用。方法选用健康Wistar大鼠32只,随机分为4组,每组8只:对照组、模型组、丹红注射液组、地奥心血康组。采用Langendorff法建立离体大鼠心肌缺血缺氧再灌注损伤模型。给药组以丹红注射液(0.33 g生药/mL)或地奥心血康灌注,直至再灌后40 min。对照组灌注KH液90 min。模型组灌注KH液35 min,全心缺血25min,再灌注30 min。测定心肌中超氧化物歧化酶、谷胱甘肽过氧化物酶、一氧化氮合酶活性变化及一氧化氮、丙二醛含量。结果超氧化物歧化酶、谷胱甘肽过氧化物酶活力:模型组明显低于其他3组(P均<0.05);丙二醛含量:模型组明显高于其他3组(P均<0.05)。一氧化氮含量:模型组明显低于其他3组(P均<0.05)。诱导型一氧化氮合酶活力:地奥心血康组明显高于模型组(P<0.05);总一氧化氮合酶活力:模型组明显高于对照组和丹红注射液组(P均<0.05),明显低于地奥心血康组(P<0.05)。结论丹红注射液对离体大鼠缺血再灌注损伤的心肌具有保护作用,此作用可能与抑制心肌脂质过氧化、增强抗氧化能力有关。

Abstract:Objective It is to observe the changes of the activities of superoxide dismutase(SOD),glutathione peroxidase,induced nitric oxide synthase(NOS) and total NOS and the contents of nitrogen monoxide and malondialdehyde(MDA) in myocardium tissue of model rats with myocardial ischemical reperfusion injury and explore the protective action of Danhong injection on ischemical reperfusion injury myocardium of rat in vitro.Methods Thirty-two healthy Wistar rats were divided randomly into control group,model group,Danhong injection group and Diaoxinxue Kang(DK) group in which there was eight rats.The rat models of myocardial ischemical reperfusion injury in vitro were established with Langendorff method.The rats in administration groups were treated with Danhong Injection(0.33 g raw drug/mL) or DK to perform perfusion for 40 min.The rats in control group were treated with KH to perform perfusion for 90 min.The rats in model group were treated with KH to perform perfusion for 35 min,total heart ischemia for 25min and reperfusion for 30 min.The changes of the activities of SOD,glutathione peroxidase and NOS in myocardium and the contents of nitrogen monoxide and MDA were determined.Results The activities of SOD and glutathione peroxidase in model group were obviously lower than those in the other three groups(P<0.05).The content of MDA in model group was obviously higher than that in the other three groups(P<0.05).The content of nitrogen monoxide in model group was obviously lower than that in the other three groups(P<0.05).The activity of induced NOS in DK group were obviously higher than that in model group(P<0.05).The activity of total NOS in model group was obviously higher than that in control group and Danhong injection group(P<0.05) and obviously lower than that in DK group(P<0.05).Conclusion Danhong injection has protective action on ischernical reperfusion injury myocardium of rats in vitro,which may be related to inhibiting myocardial lipid peroxidation and reinforcing anti-oxidizing ability.
