柚寄生总黄酮的提取工艺优化及其体外抑制人肝癌细胞Hep3B作用的初步评价

摘    要:目的:优选柚寄生总黄酮的提取工艺,并初步评价总黄酮体外抑制人肝癌细胞Hep3B的作用。方法:以芦丁为对照品测定柚寄生提取液的总黄酮含量,选取提取时间、提取温度、料液比及乙醇体积分数为影响因素,采用正交试验法优选柚寄生总黄酮的提取工艺条件,采用单因素试验考察提取次数;采用MTT法测定总黄酮对人肝癌细胞Hep3B的体外抑制作用。结果:优选的提取工艺为加30倍量60%乙醇于80℃提取2次,每次2 h;体外抗肿瘤活性试验结果表明,总黄酮质量浓度500 mg·L-1时对人肝癌细胞Hep3B的生长抑制率51.40%。结论:优选出的柚寄生总黄酮提取工艺条件稳定可行,柚寄生总黄酮有一定的体外抑制人肝癌细胞Hep3B的作用。

Abstract:Abstract Objective: to optimize the extraction process of total flavonoids from Viscum ovalifolium DC. and to evaluate the inhibitory effect of total flavonoids on Hep3B cancer cell in vitro. Method: the content of total flavonoids was analyzed by UV method, using rutin as the standard substance, and the extraction process of total flavonoids was optimized by orthogonal experiment (L9(34)). The inhibitory effect of the total flavonoids on Hep3B cancer cell in vitro was measured by MTT method. Results: the concentration of ethanol had the greatest effect on the extraction ratio of total flavonoids, and the order of the other factors that affected the extraction process was: the ratio of solid to liquid, the extraction temperature, and the extraction time. The impressive rate on the Hep3B cancer cell was 51.40% after being treated by total flavonoids at the concentration of 500 µg•mL-1. Conclusion: the optimal extraction conditions were as follows: 60% of ethanol concentration, 1:30 (g•mL-1) of solid to liquid ratio, 80 ° C of extraction temperature, 120 min of extraction time and extract twice. The total flavonoids showed some inhibitory effect on Hep3B cancer cell in vitro.
