黄芪注射液对大鼠脑缺血再灌注损伤后JNK3基因表达的影响

摘    要:目的探讨黄芪注射液对大鼠脑缺血/再灌注损伤后细胞凋亡和c-jun氨基末端激酶(JNK3)表达的影响。方法应用线栓法经左侧颈外-颈内动脉插线建立大鼠大脑中动脉阻塞再灌注(MCAO/R)模型,经腹腔注射黄芪注射液(3 mL/kg)干预治疗。Longa法评分标准评价大鼠神经行为功能,流式细胞术检测海马区神经元凋亡,Western blot检测JNK3蛋白表达,RT-PCR检测JNK3 mRNA表达。结果经黄芪注射液治疗后,大鼠海马区神经元JNK3蛋白和JNK3 mRNA表达较对照组显著减低,凋亡细胞数量显著减少,而动物神经行为功能显著改善。结论黄芪注射液可通过下调神经元JNK3基因表达而抑制细胞凋亡,改善动物的神经行为功能。

Abstract:Objective To study the effect of astragalus injection on neuronal apoptosis and expression of c-Jun N-terminal kinase3(JNK3) in cerebral ischemia reperfusion injury in rats. Methods The middle cerebral artery occlusion reperfusion rat models were established by inserting a filament suture from left external-internal carotid artery, and treated by injecting intraperitoneally astragalus injection(3 mL/kg). The neurobehavioral function was evaluated by Longa's test and the neuronal apoptosis in hippocampus detected by flow cytometry. The expressions of JNK3 mRNA and protein were respectively detected by western bloting and RT-PCR. Results After treated by astragalus injection, the expression of JNK3 mRNA and protein, the number of apoptotic neurons in hippocampus reduced significantly, while the neurobehavioral function of rats improved than those in model group. Conclusion Astragalus injection might improve the neurobehavioral function of rats by down-regulating the expressions of JNK3 and inhibiting neuronal apoptosis.
