增殖细胞核抗原与肠道损伤修复在发育生物学之间的联系

摘    要:目的研究增殖细胞核抗原(PCNA)和P53蛋白在胎儿与成人小肠组织中的表达特征。方法用免疫组化和病理技术确定这两种蛋白在胎儿小肠(19例)和成人小肠(2例)中的定位和表达量。结果在妊娠早期和中期胎儿的小肠中,PCNA蛋白存在于肠黏膜上皮细胞、固有层细胞和黏膜肌层细胞的细胞核内,表达强度为强阳性,而P53呈弱阳性表达。在妊娠晚期胎儿和成人的小肠中,PCNA为弱阳性表达,而P53表达明显升高,其阳性信号主要存在于小肠隐窝部位的细胞内。结论PCNA和P53可能通过影响细胞内DNA合成和修复,调节肠道的生长发育、结构和功能的维持以及损伤后的修复。

Abstract:Objective To explore the localization and expr ession characeteristics of prolife rating cell nuclear antigen(PCNA)and P 53 in fetal and postnatal intestines underlining their biological significance.Method The expression intensity and distribution of PCNA and P 53 were detected with pathological and immu nohistochemistry methods in 2cases of postnatal intestines and 19cases of fetal intestines with different gestational ages.Result In intestines from early and mid trim esters of pregnancy,positive signals of PCNA were mainly distributed in nu-cleuses of mucosa epithelial cells,lamina propria cells,and lamina muscular cells,while the signal of P 53 was very weak.In intestines from late trimester of pregnancy and adul t,the protein expression of PCNA was significantly decreased.P 53 content was notably increased,its protein particles were mostly located in intestinal crypt cells.Conclusion PCNA and P 53 are involved in the regulation of the intestinal de-velopment in embryonic stage,the structural and functional maintenance in adult through influencing DNA sy nthesis and repair in cells.
