壳聚糖/羟基磷灰石纳米复合材料修复骨缺损研究

摘    要:目的观察壳聚糖/羟基磷灰石纳米材料修复家兔颌骨缺损愈合的愈合过程,评价该新型材料的生物活性和成骨特性。方法电镜观测材料孔隙率、孔径大小和材料间的交连特点。30只家兔随机分为3组,分别为壳聚糖/羟基磷灰石纳米复合物充填组(A组),纳米羟基磷灰石(HA)充填组(B组)和空白对照组(C组),其中空白对照组缺损区不植入任何材料。术后2、4、8和12周分批处死家兔,取下颌骨观察缺损部愈合情况,并进行X片、力学性能的初步分析。结果壳聚糖/羟基磷灰石纳米聚合物移植10周时可完全修复骨缺损,各时段放射学评估优于其他各组,差异有显著性(P<0.05)。10周时修复效果显著差于壳聚糖/羟基磷灰石纳米聚合物充填组(P<0.05);空白组10周时缺损不能修复。骨痂力学强度与骨痂X线灰度呈指数相关(P<0.05),骨痂X线灰度随骨折愈合的进程而增高。结论壳聚糖/羟基磷灰石纳米聚合物具备良好的生物兼容性及骨引导能力,可应用此材料于骨损伤的修复,它是一种有应用前景的骨替代品和骨组织工程支架基质材料。

Abstract:Objective To observe the healing process of Chitosan/HA composites restoing rabbits jaw bones. Methods The physical and chemical properties of the composites were preliminarily studied. 30 adult rabbits were randomly divided into 3 time groups: A: Chitosan/HA group; B: HA group; C: post-operative group. After 2,4,6 and 10 weeks post-operation,the experimental rabbits were analyzed by radiographic. The animals of every group were killed according to different operalion period and the implantations were taken off and studied by gross observation and X-ray density. Results After 10 weeks post-operation,the Chitosan/HA composites group restored the wane completely,and every period X-ray estimated all good,and it had an exponential correlation (P <0.05). At 12th week HA group restore effect was worse than group A (P <0.05). The bending strength of callus and the X-ray density of callus had an exponential correlation (P <0.05). The X-ray density of callus increased coincidently with the fracture healing process. It also confirmed that there was a high correlation between the character of bending strength of callus and the X-ray density of callus. Conclusions The Chitosan/HA composites had a good biocompatibility and has some power of bone guide,it can replace homograft bone,and it is suitable for the growth of osteoblasts as well as reformation of new bone tissues.
