蜕膜组织TNFR1基因表达与原因不明早期自然流产关系探讨

摘    要:目的检测蜕膜组织肿瘤坏死因子受体1(Tumornecrosisfactorreceptor,TNFR1)的表达含量及其组织学、细胞学定位,探讨不明原因早期自然流产与患者TNFR1基因表达的关系。方法采用流式细胞仪检测技术,检测37例孕6~10周不明原因自然流产患者(流产组)和34例同期妊娠的健康妇女(对照组)蜕膜组织TNFR1的表达含量;并应用免疫组化方法,检测其中31例流产组和30例对照组蜕膜组织TNFR1表达的组织学、细胞学定位。结果(1)流式细胞仪检测流产组、对照组蜕膜组织中TNFR1的表达含量分别为(16.48±7.2)%,(12.52±5.6))%,流产组高于对照组,差异具有显著性(t=2.24,P<0.05)。免疫组化检测结果显示流产组蜕膜腺体上皮、间质、血管内皮细胞TNFR1表达率分别为77.4%,93.5%和67.7%,对照组分别为96.7%,70.0%和23.3%。2组腺体上皮细胞TNFR1表达率无显著性差异(P>0.05);流产组蜕膜间质、血管内皮细胞TNFR1表达率均高于对照组,差异均有显著性(P<0.05,P<0.05)。结论蜕膜组织中TNFR1表达水平升高,主要分布在蜕膜间质和血管内皮细胞表面,这种改变可能与不明原因早期自然流产有关。

Abstract:Objective: To investigate the expression and histological localization of tumor necrosis factor receptor 1 (TNFR1) in deciduas of unexplained early spontaneous abortion. Methods: 37 women with unexplained spontaneous abortion at 6~10 weeks of gestational period servedas abortion group. The control group consisted of 34 normal pregnancy women who preferred artificial abortion at the same gestational period. Decidua samples from the two groups were taken at the time of abortion. The flow cytometer was used to detect the level of TNFR1 expressed in deciduas. The immunohitochemisty was used to determine the histological localization of TNFR1 stainning in 31 abortion and 30 control group deciduas. Results: (1) The percentage of TNFR1 positive decidual cells was (16.48±7.2) % for abortion group , it was significantly higher than control group ( 12.52±5.6)%,t=2.24, P<0.05). (2) the rate of TNFR1 expressed in decidual glandular epithelia cell, stromal cell and blood vessel endothelial cell of abortion group were 77.4%, 93.5% and 67.7% respectively, and in control groups 96.7%, 70.0% and 23.3% respectively. There was no significant difference of TNFR1 expressive rate in decidual glandular epithelia cell between the two groups (P >0.05 ),however , the expressive rate of TNFR1 in decidual stromal cell and blood vessel endothelia cell from the abortion group was higher than that from control group (P<0.05, P<0.05).
