山西产葛根总黄酮及3种异黄酮含量的测定

摘    要:目的:测定山西产葛根总黄酮及3种异黄酮的含量。方法:紫外分光光度法测定葛根总黄酮的含量,检测波长250nm。高效液相色谱法测定葛根3种异黄酮的含量,色谱柱为Uvis-201(4.6mm×250mm,5μm),流动相为甲醇(A)-水(B)梯度洗脱,流速为1.0mL/min,柱温为室温;检测波长250nm。结果:葛根总黄酮在4.32-21.60μg/mL范围内线性关系良好,平均回收率为99.20%,RSD为1.57%;葛根素、大豆苷、大豆苷元的进样量分别在0.2124-4.2480μg、0.1207-2.4140μg、0.0883-1.7660μg范围内呈良好的线性关系,平均回收率分别为99.96%、99.02%、99.12%,RSD分别为1.68%、2.50%、2.19%。结论:本方法简单准确,重复性好,适用于葛根总黄酮及3种异黄酮的含量测定。

Abstract:Objective: To determine the contents of total flavonoids and three isoflavonoids in Pueraria lobata.Methods: The ultraviolet-visible spectrophotometry was used for the determination of total flavonoids in Pueraria lobata at the detection wavelength of 250nm.The high-performance liquid chromatography was adopted for the determination of three isoflavonoids in Pueraria lobata.Chromatographic column: Uvis-201 column(4.6mm×250mm,5μm);mobile phase: methanol(A)-water(B)(gradient elution);flow rate: 1.0mL/min;column temperature: ambient temperature;detection wavelength: 250nm.Results: The linear relation of total flavonoids was good in the range of 4.32-21.60μg/mL.The average recovery rate was 99.20%,RSD was 1.57%.The linear relations of puerarin,daidzin and daidzein were good in the range of 0.2124-4.2480μg,0.1207-2.4140μg,and 0.0883-1.7660μg respectively.The average recovery was 99.96%,99.02%,99.12%;RSD was 1.68%,2.50%,2.19% respectively.Conclusion: This method is simple and accurate in operation and good in repeat.It is suitable for the determination of total flavonoids and three isoflavonoids in Pueraria lobata.
