硫酸软骨素胰管注射对减轻大鼠急性坏死性胰腺炎胰腺细胞损伤的作用

摘    要:目的:探讨硫酸软骨素胰管注射减轻大鼠急性坏死性胰腺炎(ANP)胰腺组织氧化应激损伤及对胰腺细胞的保护作用.方法:雄性Wistar大鼠(n=36)随机分3组,A组为ANP模型组(5%牛磺胆酸钠逆行胰管注射);B组为ANP诱导1 h后硫酸软骨素处理组;C组为假手术组.3组动物分别于6 h处死.检测血清淀粉酶(SAM)、胰腺组织中丙二醛(MDA)、三磷酸腺苷(ATP)、还原型谷胱苷肽(GSH)、超氧化物歧化酶(SOD)含量,并观察胰腺组织细胞形态学改变.结果:A、B两组均有SAM水平的升高,病理组织学上有相应的ANP变化.A组胰腺组织中ATP、GSH、SOD明显下降(P<0.01)、B组则下降幅度较小,与A组比较差异有显著性(P<0.05);A组MDA明显升高(P<0.01),B组升高幅度小,与A组比较差异有显著性(P<0.05)结论:ANP时胰腺组织中内源性抗氧化物质显著下降,脂质过氧化增加,能量耗竭.胰、胆管注射硫酸软骨素可作为氧自由基清除剂明显减轻ANP大鼠胰腺细胞损伤.

Abstract:Objective: To explore the effect of chondroitin sulfate (CS) injected via the pancreatic duct on the oxidative stress damage of pancreatic tissue and its protective effect on pancreatic cell in rats with acute necrotic pancreatitis (ANP). Methods:Thirty-six male Wistar rats were randomly divided into 3 groups: group A (ANP model group), group B (CS treatment group), and group C (control group). Rats in these 3 groups were killed 6 hours later. The levels of serum amylase (SAM), malonyldialdehyde (MDA), adenosine triphosphate (ATP), glutathione (GSH), and superoxide dismutase (SOD) were measured. Histopathological changes were observed. Results:The level of SAM increased in both group A and group B, but in group A it was significantly higher. The levels of ATP, GSH, and SOD in group B were significantly higher than those in group A ( P< 0.05), but the level of MDA in group B was significantly lower than that in group A ( P< 0.01). Conclusion:Chondroitin sulfate injected via pancreatic duct could serve as the scavenger of oxygen free radicals and thus obviously reduce the pancreatic cell damage in ANP rats.
