缺氧条件下硒对大鼠血管活性物质的效力

摘    要:目的研究不同剂量硒在缺氧条件下对大鼠血管三种活性物质的效力。方法随机将成年SD雄性大鼠分为正常对照组、缺氧对照组、缺氧加硒1-4组。缺氧对照组和缺氧加硒组在缺氧装置中给予8%氧气,2 h/d,饲料中添加不同剂量硒,喂养30 d后取血,采用酶联免疫法(ELISA)测定血清中三种血管活性物质的量。结果三种活性物质缺氧加硒组与缺氧对照组比较,差异均有统计学意义(P〈0.01)。5倍和20倍剂量硒组降低ANG-Ⅱ的活性效力明显;5倍和40倍剂量硒组降低内皮素-1(ET-1)的活性效力明显;40倍剂量硒组增加TNF-α的活性效力明显。结论缺氧条件下,硒浓度变化对大鼠ANG-Ⅱ、ET-1、TNF-α的效力有影响。提示采用改变硒浓度的方式可改善缺氧对心血管系统的不良影响。

Abstract:To investigate the efficacy of selenium on three kind of vascular active substances under hypoxia environment in rats. Methods Adult male SD rats were randomly divided into normal control group, hypoxia group and hypoxia plus selenium( 1 -4)group. 8% oxygen in device were given for hypoxia control group and plus selenium group 2h/d, and different doses selenium were fed. Taken blood after 30 days, enzyme - linked immunoas- say (ELISA) had determined amount of three kinds of vascular active substances. Results Three kinds of active sub- stances in hypoxia plus selenium group showed statistical significant ( P 〈 0.01 ) compared with hypoxia control group. Selenium at 5 and 20 times dose could significantly reduce ANG - Ⅱ activity;selenium at 5 and 40 times dose could reduce ET - 1 activity as well ; selenium at 40 times dose could significantly increase TNF - α activity. Conclusion Under the hypoxia environment, different concentration of selenium can influence rats' ANG - Ⅱ, ET - 1 and TNF - α activity and it indicate that changes of selenium concentration can improve the negative effects on cardiovascular system.
