摘    要:目的:检测RUNX3、SMAD4和VEGF蛋白在胃腺癌组织中的表达,观察其与胃腺癌临床病理因素的关系,探讨RUNX3、SMAD4基因在胃腺癌浸润、转移过程中的作用及意义。方法:收集病人临床资料,免疫组织化学SP法检测97例胃腺癌及20例正常胃黏膜组织中RUNX3、SMAD4和VEGF的表达情况。SPSS 13.0统计软件进行数据处理3指标表达与临床病理参数之间的关系,及3项指标之间的相关性。结果:①胃腺癌组织中RUNX3、SMAD4阳性表达率明显低于它们在正常胃黏膜组织中的阳性表达;VEGF在胃腺癌组织中的阳性表达率明显高于正常胃黏膜组织;上述表达差异有显著性。②RUNX3,SMAD4低表达,VEGF高表达与胃腺癌分化程度、浸润深度、淋巴结转移相关,差异具有显著性。③胃腺癌组织RUNX3与SMAD4阳性表达率之间呈正相关,二指标与VEGF阳性表达率均呈负相关,差异具有显著意义。结论:胃腺癌组织中RUNX3、SMAD4低表达可能协同上调VEGF的表达,从而促进胃癌的浸润、转移;RUNX3、SMAD4、VEGF可以作为判断胃癌预后及指导临床治疗的指标。

Abstract:Objective: To detect the protein expressions of RUNX3,SMAD4,and VEGF in human gastric cancer tissue and explore their significance on invasion and metastasis of gastric carcinoma.Methods: The expressions of RUNX3,SMAD4,and VEGF were detected by immunohistochemistry in 97 cases of gastric carcinoma and 20 subjects with normal gastric mucosa.Results: The positive rates of RUNX3,SMAD4,and VEGF in gastric cancer tissues were 56.7%,54.6%,76.3% respectively,while in 20 normal tissues were 90%,83.3%,56.7% respectively,with significant difference between the two groups(P<0.05);the expressions of RUNX3,SMAD4,and VEGF were closely correlated with gastric cancer differentiation,depth of invasion,and lymph node metastasis(all P<0.05).In gastric carcinoma tissues,RUNX3 expression was positively correlated with SMAD4,but RUNX3 and SMAD4 were both inversely correlated with the expression of VEGF.Conclusion: RUNX3 may have synergetic effects with SMAD4 the invasion and metastasis through regulating VEGF expression.The detection of RUNX3,SMAD4,and VEGF may be helpful for evaluation of the prognosis of the patient with gastric carcinoma.
