表没食子儿茶素没食子酸脂对前列腺癌PC-3细胞体外生长的抑制作用

目的:研究表没食子儿茶素没食子酸脂(EGCG)对人前列腺癌细胞体外生长的抑制作用,以及其对细胞增殖周期的影响。方法:用不同浓度的EGCG处理体外生长的前列腺癌PC-3细胞,通过对细胞形态的对比观察和MTT法检测EGCG对PC-3细胞的抑制作用,通过流式细胞术检测其对细胞周期的影响。结果:与对照组相比,EGCG处理组可使细胞明显变圆,体积增大,细胞内颗粒及脱壁细胞增多;MTT法显示,EGCG能明显抑制前列腺癌PC-3细胞生长,并呈时间、剂量依赖性(P<0.05);流式细胞术显示,EGCG处理的PC-3细胞被阻止在G0/G1期,并有凋亡峰出现。结论:EGCG通过阻滞细胞的增殖周期对前列腺癌PC-3细胞体外生长有明显的抑制作用。

Inhibitive Effects of EGCG on the Proliferation of Human Prostate Cancer Cell Line PC-3

Objective: To study the inhibitive effect of(-)-epigallocatechin gallate(EGCG) on the proliferation and cell cycle of human prostate cancer cells in vitro.Methods: The morphological alterations of PC-3 cells treated by EGCG were observed under inverse microscopy.The cell proliferation was assayed by MTT method,and the cell cycle changes were observed by flowcytometry.Results: After being treated with EGCG,PC-3 cells were round and small,and grew against the wall under inverse microscope.In comparison to the control,all the different concentrations of EGCG were able to inhibit the growth of malignancy cell lines,and the inhibition of proliferation of PC-3 cells increased in a time-and dose-dependent manner(P<0.05).Furthermore,the cell cycle of PC-3 cells treated by EGCG was arrested in the G0/G1 phase in a dose-dependent manner.Conclusion: That EGCG has significantly inhibitory effect on the PC-3 cells in vitro through arresting the cell cycle.
