塞来昔布对裸鼠人肝癌细胞株SMMC-7721移植瘤增殖的影响

摘    要:目的 探讨环氧化酶-2(COX-2)抑制剂塞来昔布对裸鼠人肝癌细胞皮下移植瘤生长抑制作用及其相关机制.方法 用人原发性肝癌细胞株SMMC-7721接种BALB/C(nu/nu)裸鼠背部皮下,建立裸鼠人肝癌细胞皮下移植瘤动物模型(n=20)并分成实验组(n=10)和空白对照组(n=10).实验组接种前3 d开始在饮水中添加塞来昔布,对照组给予正常饮水,观察肿瘤生长曲线和质量,并应用免疫组化法对肿瘤组织的细胞凋亡情况、血管内皮生长因子(VEGF)和肿瘤微血管密度(MVD)表达程度进行观察.结果 实验组肿瘤出现的时间显著晚于对照组,肿瘤的体积、质量显著小于对照组,两组差异有高度统计学意义(P<0.01);塞来昔布处理后肿瘤组织的细胞凋亡明显强于对照组,而VEGF、MVD表达明显弱于对照组.结论 COX-2抑制剂塞来昔布可通过诱导凋亡及抑制肿瘤新生血管生成而抑制裸鼠人肝癌细胞SMMC-7721株移植瘤的生长,其作用途径是抑制了COX-2的表达.
