骨髓间充质干细胞移植大鼠脑梗死区的超微结构变化

摘    要:背景近来的研究发现骨髓间充质干细胞(mesenchymal stem cells,MSCs)对缺血性脑血管病有治疗作用,接受细胞移植的脑梗死区超微结构的观察可进一步证实该作用.目的观察接受MSCs局部注射移植的大鼠脑梗死区超微结构的变化,以了解MSCs移植对梗死区超微结构的影响.设计随机对照实验研究.单位解放军第三军医大学西南医院神经病学中心.材料实验于2002-04/2002-09在解放军第三军医大学动物所及中心实验室完成.实验动物为健康幼年及成年Wistar大鼠12只,雌雄不限.随机分为移植组6只和对照组6只.干预建立大鼠大脑中动脉栓塞模型,将体外分离培养的大鼠MSCs注射至大鼠脑梗死区,两组大鼠于移植后第4周电镜观察大鼠脑梗死区超微结构变化.主要观察指标移植组及对照组大鼠脑梗死区超微结构变化.结果接受移植的梗死区可见外形幼稚的细胞存活,并围绕在神经元周围,梗死区神经元胞浆中出现了丰富的游离核糖体,而对照组大鼠梗死区神经元胞核固缩,并有嗜神经细胞现象.结论大鼠MSCs移植对缺血性脑损伤具有一定修复作用.

Changes of ultrastructure in cerebral infarction region of rats with transplantation mesenchymal stem cells
Abstract:BACKGROUND: It is recently found that mesenchymal stem cells(MSCs)have therapeutic function for ischemic cerebrovascular disease and that the function should have a further observation of the ultrastructural changes in cerebral infarction region with quantitative MSCs transplantation.OBJECTIVE: To observe the ultrastructural changes in cerebral infarction region of rats injected locally with mesenchymal stem cells so as to understand the effect of MSCs transplantation on the ultrastructure in cerebral infarction region.DESIGN: A randomized and controlled trial.SETTING: Neurology Center of Southwest Hospital Affiliated to the Third Military Medical University of Chinese PLA.MATERIALS: The experiment was done in the medical Animal Institute and Central Laboratory of the Third Military Medical University of Chinese PLA from April 2002 to September 2002. The experimental animals were 12 young and adult healthy Wistar rats, without limitation of sexes. They were randomly divided into two groups: transplantation group and control group with six rats in each group.INTERVENTIONS: The model of rats with embolism in middle cerebral artery was established. MSCs of rats separately cultured in vitro were injected in cerebral infarction region of rats. Changes of ultrastructure in cerebral infarction region of rats were observed by electron microscope in the 4th week after transplantation in the two groups.MAIN OUTCOME MEASURES: Changes of ultrastructure in cerebral infarction region in rats of transplantation group and control group.RESULTS: Cells with immature outline were survived in infarction region after transplantation and around the neurons. Plenty of free ribosomes were found in neuronal cytoplasm of infarction region. But in the control group,pycnosis of neuron and neurotropic cells were found in infarction region.CONCLUSION: MSCs transplantation has certain repair function for the ischemic cerebral injury.
