| 摘 要： | 目的观察中介素1-53(IMD1-53)对大鼠肺缺血再灌注损伤(LIRI)的保护作用及其可能的机制。方法建立大鼠在体肺缺血再灌注模型。将健康雄性Wistar大鼠72只随机分为手术对照组(C组)、缺血再灌注组(IR组)、IMD1-53干预组(D组)。每组分别在缺血45min,再灌注60、120min时处死8只大鼠,取血浆和右肺中叶,测定血清丙二醛(MDA)含量、髓过氧化物酶(MPO)活性;反转录聚合酶链反应(RT-PCR)测定肺组织IMD、降钙素受体样受体(CL)mRNA水平;放射免疫法测定血浆IMD1-53含量,并进行肺组织病理学检查。结果与正常对照组相比,再灌注期间IR组血浆MDA含量、肺组织MPO活性升高,肺组织IMD、CL基因表达水平及血浆中IMD1-53含量显著降低,而IMD1-53干预后可以减弱缺血再灌注诱导上述指标的变化,肺组织病理学检查显示肺缺血再灌注损伤程度也较IR组明显减轻。结论IMD1-53具有抗氧化作用,可抑制氧自由基生成、中性粒细胞浸润,减少脂质过氧化作用,对肺缺血再灌注损伤有保护作用。
