戊四氮诱发大鼠全身强直-阵挛性癫痫发作诱导终纹床核c-fos癌基因表达

摘    要:本文用Fos癌蛋白抗体免疫组化法详细研究了戊四氮诱发大鼠全身强直-阵挛性癫痫发作诱导终纹床核c-fos癌基因表达的动态分布,结果如下:腹腔内注入戊四氮50mg/kg后0.5h,前腹区梭形核出现低密度的Fos免疫阳性(Fos-ir)细胞核标记;注射戊四氮后3h,前腹区和后部腹外侧区域均有较高密度的Fos-ir标记,前外区和前背区的密度则较低,后部背内侧区域未见Fos-ir标记;注射戊四氮后6h,终纹床核各核团和区域均出现较高密度的Fos-ir标记,其中,前部的密度高于后部、腹侧区域的密度高于背侧区域。上述终纹床核的Fos-ir标记出现的先后顺序及波及范围很可能反映了戊四氮诱发大鼠全身强直-阵挛性癫痫发作累及终纹床核的顺序和范围。

Abstract:By using Fos-oncoprotein antibody immunohistochemieal technique, the changes of distribution of c-los oncogene expression within the bed nuclei of the stria terminalis (BST) in pentylenetetrazol (PTZ)-induced systemic tonic-clonic epilepsy in the rat was studied in detail. The results are as followst 30 minutes following PTZ injection (50 mg/kg, i. p. ), a small number of Fos-immunoreactive (Fos-ir)cell nuclei appeared in the fusiform nucleus of the anteroventral area; 3 hours after PTZ injection, the anteroventral area and ventrolateral region of the posterior part contained relatively high density of Fos-ir labeling, while relatively low density of it was displayed in the anterolateral and anterodorsal areas, and at the same time, no Fos-ir labeling could be found in the dorsomedial region of the posterior part; after another 3 hours, highly concentrated Fos ir cells was distributed in each nucleus and region of the BST, with higher density in the anterior part and ventral region than in the posterior part and dorsal region. The above mentioned relation between time and involving extent of the BST of Fos-ir labeling may well reflect the spreading sequence and range in BST of PTZ-induced systemic tonic-clonic epileptic attacks.
