摘    要:目的研究输注转染人IL-10基因的骨髓间充质干细胞(hIL-10-MSC)对大鼠原位异种肝移植后移植肝的保护作用及机制。方法建立豚鼠对Wistar大鼠的非协调性异种原位肝移植模型,受体分为3组:空白对照组、骨髓间充质干细胞(MSC)对照组、hIL-10-MSC组。术后12h取移植肝,HE染色观察移植肝组织形态学变化,用RT-PCR、蛋白质印迹法观察移植肝组织IL-10、穿孔素、颗粒酶B的表达情况。结果与空白对照组、MSC对照组相比,hIL-10-MSC组大鼠移植肝排斥反应最轻,穿孔素、颗粒酶B的表达明显降低,而IL-10的表达显著升高(均P〈0.05)。结论 hIL-10-MSC对移植肝的保护作用可能与其减少穿孔素/颗粒酶B的表达有关。

Abstract:Objective To investigate the protective role of bone marrow mesenchymal stem cells(MSC) transfected with human IL-10 gene in rat orthotopic xenogeneic liver transplantation.Methods Model of cavia to rat discordant liver xenotransplantiaion was set up.The recipients were randomly divided into 3 groups:control group,MSC group,and hIL-10-MSC group.The transplanted rats were sacrificed 12 h after operation and the liver grafts were taken out for morphological observation with HE staining and assessing expressions of IL-10,perforin,and granzyme B with RT-PCR and Western blotting.Results Immunological rejection of the hIL-10-MSC group was milder than that of the control and MSC groups.Compared with the control and MSC groups,the expression of perforin and granzyme in the hIL-10-MSC group was significantly decreased,while the expression of IL-10 was dramatically increased(all P<0.05).Conclusion hIL-10-MSC may play a protective role in liver graft function by diminishing perforin and granzyme B.
