人恶性胰岛细胞瘤SCID鼠原位移植瘤免疫组织化学和超微结构的研究

摘    要:目的 为探讨人恶性胰岛细胞瘤转移机制和抗转移治疗提供实验工具。方法 将人恶性胰岛细胞瘤组织植入SCID胰腺被膜下 ,观察移植瘤的侵袭转移及形态学特征 (光镜、免疫组织化学、电镜 )。结果 在SCID鼠体内成功地建立了一株人恶性胰岛素瘤原位移植模型HMI HMN 1,已传 2 1代 ,和一株人恶性胰多肽瘤原位移植模型HPPT HMN 2 ,已传2 5代 ,共移植SCID鼠共 14 7只 ,其移植生长率和自发转移率达 10 0 %。表现为肿瘤广泛侵袭胃十二指肠并有淋巴结和肝转移。病理组织学、免疫组织化学和电镜观察结果证明移植瘤细胞与瘤源人胰岛细胞瘤细胞完全一致。结论 人恶性胰岛细胞瘤SCID鼠原位移植模型的建立为研究人恶性胰岛细胞瘤转移机制和抗转移治疗提供了理想的动物模型。

Abstract:Purpose To provide an experimental model for the research of human malignant tumors of pancreas islet cells. Methods Orthotopicly transplanted the human tumor tissues into the cover membrane of pancreas in SCID mice, and its invasion metastasis and morphologic characteristics (light microscopy, electronic microscopy, immynohistochemistry) were observed. Results A model of human malignant insulinoma(HMI HMN 1) was successfully established in SCID mice, which passaged 21 generations, and a model of human malignant polypeptide tumor(IIPPT IIMN 2), which passaged 25 generations. Both the growth rate and spontaneous metastasis rate were 100% in 147 SCID mice, in which diffusely invasion of stomach and duonenum, lymph nodes and liver metastasis were present. The tranplatation tumor cells had the consistency to the original tumors of human pancreas islet cells. Conclusion The two models of human malignant tumor of pancreas islet cells in SCID mice are ideal for the research of the metastatic mechanism and therapy.
