P16基因蛋白在胃癌中的表达及意义

目的检测胃癌及慢性萎缩性胃炎、慢性浅表性胃炎患者的P16的表达水平，探讨抑癌基因在胃癌发生中的作用。方法采用Envisoon二部法对93例胃镜下胃黏膜组织石蜡切片滴加P16单克隆抗体并染色。每批染色均设阴性对照，用PBS替代一抗。用已知阳性切片为阳性对照。免疫组化显色判断采用许氏阳性细胞半定量法^[2]0判定P16的阳性率。结果P16阳性率：慢性浅表性胃炎组为50％（15／30），萎缩性胃炎组为68．8％（21／32），不典型增生组58％（7／12），胃癌组29％（9／31），所得数据X^2检验，胃癌组和慢性浅表性胃炎组者比较二者差异无统计学意义（P〉0．01）。胃癌和不典型增生组间比较二者差异无统计学意义（P〉0．01）。胃癌和萎缩性胃炎组比较二者差异无统计学意义（P〈0．05）。结论P16蛋白在胃癌的发生发展中起着明显的作用，对预后有很高的价值。

The expression and significance of P16 protein gene in gastric carcinoma

Objective To investigate the expression standard in patients of gastric carcinoma,chronic atrophic gastric and chronic superficial gastric,to discuss the function of carcinoma repressor gene in gastric carcinoma.Methods Adopt by Envision TM staining,the gastric tissues in paraffin-embedded were effected on P16 specific monoclonal antibodies and the other were composed of PBS.P16 protein gene were characterized by semi-quantitative of immunohistochemistry.Results Immunohistochenistry analysis showed that P16 positive rate:50%(15/30)in chronic superficial gastric group,68.8%(21/32)in chronic atrophic gastric group,58% (7/12)in dysplasia group and29%(9/31)in gastric carcinoma group.There was no difference in comparison of gastric(P＞0.01),and there was no difference in comparison of gastric carcinoma and dysplasia(P＞0.01),string difference was found in comparison of gastric carcinoma and chronic atrophic gastric(P＜0.05).Conclusion P16 Protein gene may play an important role in the pathogenesis in gastric carcinoma.
