一氧化氮与慢性胃溃疡关系的实验研究

摘    要:采用乙酸注射至大鼠胃窦部浆膜下形成慢性胃溃疡模型 ,用游标卡尺测定溃疡指数 ,用 SABC免疫组织化学的方法 ,观察表皮生长因子受体的表达情况并进行图像分析 ,用镀铜镉还原比色法测定血清 NO3- / NO2 - 含量代表血清一氧化氮水平 ,以研究一氧化氮与慢性胃溃疡之间的关系 ,了解在胃溃疡愈合过程中表皮生长因子受体在溃疡周围粘膜的表达 ,探索它们在大鼠慢性胃溃疡的病理生理意义。结果 :1L- Arg组溃疡指数显著低于模型组和 L- NNA组 (P<0 .0 5 ) ,L- NNA组溃疡指数显著高于模型组 (P<0 .0 5 )和 L- Arg组 (P<0 .0 1)。 2模型组和 L- Arg组表皮生长因子受体表达较对照组增加 (积分光密度值及阳性细胞占总面积的百分比均为 P<0 .0 1) ,L - NNA组与对照组和模型组相比无显著性差异 (积分光密度值和阳性细胞占总面积的百分比均为 P>0 .0 5 )。3L - Arg组血清 NO3- / NO2 - 含量显著高于模型组 (P<0 .0 1)及对照组 (P<0 .0 1) ,L - NNA组血清 NO3- / NO2 - 含量显著低于其余 3组。认为正常一氧化氮水平有利于慢性胃溃疡的愈合 ,一氧化氮对表皮生长因子受体的表达具有重要意义 ,这可能是一氧化氮促进慢性胃溃疡愈合的机理之一。

Abstract:The relationship between nitric oxide (NO) and chronic gastric ulcer in rats and the changes and possible role of the expression of epidermal growth factor receptor (EGFR) around the ulcer were investigated. The rat model of chronic gastric ulcer was developed by injection of acetic acid into the submucous layer. Wistar rats ( n =34) were divided into control group ( n =9), model group ( n =8), L Arg treated group ( n =9) and L NNA treated group ( n =8). The index of ulcer (UI) was measured by using a vernier, EGFR receptor expression detected by immunohistochemical method (SABC) and underwent micro image analysis, and serum NO 3 -/NO 2 - representing serum NO level determined by reduced chromatometry. The results were as follows: (1) UI in L Arg group was significantly lower than in model group and L NNA treated group ( P <0.05), while UI in L NNA group obviously higher than in model group ( P <0.05) and L Arg treated group ( P <0.01). (2) The expression of EGFR in model group and L Arg group was increased as compared with that in the control group ( P <0.01),but there was no significant difference between L NNA treated group and control group or model group ( P >0.05). (3) The level of serum NO 3 -/NO 2 - in L Arg treated group was dramatically higher than in model group and control group ( both P <0 01), while significantly lower than in the other three groups. It was suggested that NO could accelerates gastric ulcer healing due to the increase of EGFR expression by NO.
