兔角膜碱烧伤后角膜组织中G-6-PD和胶原酶活性变化

摘    要:目的:探讨角膜组织中6 - 磷酸葡萄糖脱氢酶(G- 6 - PD) 和胶原酶活性变化与角膜碱烧伤的关系。方法:用2mol/LNaOH溶液造成右眼角膜碱烧伤模型,以左眼为正常对照。分别于烧伤后不同时期测定两组角膜中的G- 6 - PD 的活性和氧化型谷胱甘肽(GSSG) 及羟脯氨酸的含量。结果:与对照组相比,在烧伤后的1 - 16 天,实验组的G- 6 - PD活性和羟脯氨酸含量下降,而GSSG 含量升高,尤其是第8 天的差异非常显著(P< 0 .01) ,至第32 天时上述指标基本接近对照组水平。G- 6 - PD 活性与羟脯氨酸含量呈正相关,而与GSSG 含量呈负相关。结论:G- 6 - PD活性降低导致GSSG 含量升高继而胶原酶活性增强,可能是角膜碱烧伤的机制之一。

Abstract:Objective To explore the relation between the corneal alkali burn and the change of Glucose-6-phosphate dehydrogenase (G-6-PD)and collagenase.Metheds The model of corneal burn was established with 2mol/L NaOH solution in the right eyes of rabbits while the left eyes as control group,and then measure the G-6-PD activity and the content of hydroxyprolin and oxidized glutathione(GSSG) in cornea tissue at different time after alkali burn.Results From the 1st to 16th days after the burning,the G-6-PD activity and hydroxyprolin content of experimental group decreased but GSSG increased,these indexes was found markedly different than those of control group (P<0.01) especially on the 8th day.On the 32nd day,the above indexes returned to the normal level.There was significant positive correlation between G-6-PD activity and hydroxyprolin content,but negative correlation between G-6-PD activity and GSSG content.Conclusion It is possibly one of the machanism of corneal alkali burn that lowered G-6-PD activity results in increasing of GSSG content and following activation of collagen ase.
