基因芯片在肾癌相关基因研究中的应用

摘    要:目的 :通过研究人肾癌组织和癌旁正常组织中差异表达的基因 ,寻找肾癌相关基因以用于诊断和治疗。方法 :以包含8000个cDNA基因表达谱芯片研究一组肾癌组织样本的基因表达谱。按一步法抽提肾癌和对照组癌旁正常组织的总RNA并纯化mRNA ;将等量的对照组组织和肾癌组织mRNA分别逆转录合成荧光分子掺入的cDNA—链做探针 ,混合后杂交上述基因芯片。经严格洗片后用ScanArray3000扫描仪扫描芯片荧光信号图像 ,计算后分析比较两种组织中差异表达的基因。结果 :共筛选出差异表达基因95条 ,其中未知基因44条。结论 :基因芯片在筛选肾癌相关基因的改变具有快速、高通量、灵敏的特点 ,肾癌在细胞代谢、信号转录、蛋白合成的相关基因方面有异常表达

Abstract:Objective:To investigate the genes differentially expressed in renal carcinoma and screen renal cell carcinoma associated genes.Methods:The cDNA retro-transcribed from equal quantity mRNA from renal cell carcinoma and adjacent normal renal tissues,which were labeled with cy5 and cy3 fluorescence as probes.The mixed probe was hybridized with two piece H80 gene chip.It was scanned by laser scanner.The acquired image was analysed software.Results:A total of 1333 genes were screened out which expression level were more than 2 times in renal cell carcinoma and adjactive normal renal tissue.A total 44 new genes were found in the study.Conclusion:The cDNA microarray for analysis of gene expression profile is a powerful method to identify renal cell carcinoma associated genes,which involved to cell cycle,signal transduction,protein synthesis and metablism.
