结核病患者外周血B细胞及其亚群的检测

目的初步建立B细胞分群技术,并了解健康对照者、结核潜伏感染者、结核患者B细胞亚群的分布情况。方法根据B细胞表面CD19/CD5/CD1d标识将B细胞分为四群。利用流式细胞技术比较健康对照、结核潜伏感染者、结核患者外周血B细胞及B细胞分群表达的差异。结果结核患者外周血B细胞较健康对照者减少,但结果无统计学差异。结核患者外周血CD19+CD5+CD1d+B细胞显著高于健康对照及结核潜伏感染者(P<0.05);CD19+CD5-CD1d+B细胞高于结核潜伏感染者(P<0.05);CD19+CD5-CD1d-B细胞低于结核潜伏感染者(P<0.05)。结论肺结核患者外周血中存在着B细胞亚群的紊乱,尤其是CD19+CD5+CD1d+B细胞增多最为明显,而CD19+CD5-CD1d+B/CD19+CD5-CD1d-B细胞只比结核潜伏感染者显著增多或降低,提示B细胞亚群在结核的发病中起重要作用。

Analysis of B cells and B cell subsets in the peripheral blood of patients with pulmonary tuberculosis

Objective The initial establishment of B-cell clustering techniques,and understanding the distribution of B cell subsets among the healthy controls,latent tuberculosis infection and the TB patients.Methods According to the CD19/CD5/CD1d on the surface of cell,the B cells are divided into four groups.The B cell subsets of peripheral blood among the healthy controls,latent tuberculosis infection and the TB patients were analyzing with flow cytomety.Results The frequency of the B cells in TB patients was lower than HD,but the result was no significant difference.The proportion of CD19+CD5+CD1d+B cells in TB patients was much more than HD and LTBI(P<0.05),That of CD19+CD5-CD1d+B was higher than LTBI(P<0.05).The CD19+CD5-CD1d-B cells in TB were decreased compared with LTBI(P<0.05).Conclusion The B cell subsets were confused in peripheral blood of patients with pulmonary tuberculosis.In particularly,CD19+CD5+CD1d+B cells obviously increased.Compared with LTBI,CD19+CD5-CD1d+/CD19+CD5-CD1d-B cells increased or decreased.
