人caveolin-1基因慢病毒过表达载体构建及其对SMMC7721细胞凋亡的影响

摘    要:目的 构建人caveolin-1基因慢病毒过表达载体,并研究其对人肝细胞癌株SMMC7721细胞凋亡的影响. 方法 应用慢病毒载体系统构建人caveolin-1基因pGC-FU-caveolin-1过表达质粒,并转导SMMC7721细胞,检测其转导效率、细胞内caveolin-1 mRNA和蛋白水平的改变和对细胞凋亡的影响. 结果 pGC-FU-caveolin-1过表达质粒慢病毒转导SMMC7721细胞72 h的转导效率为(96.3±1.6)%.转导72 h和120 h后caveolin-1 mRNA水平分别是阴性慢病毒感染组的(30.15±1.22)倍和(189.16±28.13)倍,两个时相点的差别均具有统计学意义(P<0.05).同期转导后120 h的蛋白水平检测明显升高.pGC-FU-caveolin-1过表达质粒慢病毒转导SMMC7721细胞120 h后能使早期细胞凋亡数减少约70%,两者差别具有统计学意义(P<0.05). 结论 成功构建的pGC-FU-caveolin-1过表达质粒慢病毒能在SMMC7721细胞中长期稳定表达,其过表达能显著抑制SMMC7721细胞凋亡.
