隔药饼灸对子宫内膜异位症大鼠雌激素相关因子表达的影响

摘    要:目的:探索隔药饼灸对子宫内膜异位症大鼠雌激素相关因子表达的影响。方法:采用自体移植法造模,成功后随机将SD大鼠分为模型组、隔药饼灸组和达那唑组,另设未造模的大鼠为空白对照组。空白对照组和模型组大鼠常规固定、暴露关元穴,不做其他处理;其余两组分别给予隔药饼灸法治疗、达那唑悬浊液灌胃,共2个疗程。待治疗结束后,用HE染色观察子宫内膜组织及异位内膜组织形态学变化;ELISA法检测血清E2水平;RT-PCR检测内膜组织芳香化酶(P450arom)和环氧化酶-2(COX-2)的mRNA表达。结果:HE染色结果显示,模型组的异位内膜上皮细胞呈柱状增生,间质细胞密集,局部纤维化,血供丰富;隔药饼灸组的异位内膜上皮细胞萎缩,呈矮立方状,间质细胞萎缩,血供减少,局部呈纤维化。与空白对照组比较,模型组大鼠血清E2水平和异位内膜组织P450arom mRNA、COX-2 mRNA的表达量显著升高(P0.05);与模型组比较,隔药饼灸组和达那唑组大鼠血清E2水平和异位内膜组织P450arom mRNA、COX-2的mRNA表达量明显降低(P0.05);与隔药饼灸组比较,达那唑组血清E2水平和异位内膜组织P450arom mRNA、COX-2 mRNA表达量明显降低(P0.05)。结论:隔药饼灸可能通过改善血液循环,降低子宫内膜异位症大鼠血清E2和异位内膜组织中P450arom、COX-2的表达,从而抑制雌激素的生成代谢、抑制异位病灶的生长。

Abstract:  Objective:To explore the effects of medicinal cake-separated moxibustion on the expressions of estrogen-related factors in rats with endometriosis.   Methods: The model was established by autogenous transplantation. After successful modeling,SD rats were randomly divided into the model group,medicinal cake-separated moxibustion group and danazol group. Additionally,the normal rats without modeling were taken as the blank control group. The rats in the blank control group and model group were routinely fixed and the Guanyuan(CV4)acupoint was exposed,without other treatment. The medicinal cake-separated moxibustion group was treated with the therapy of medicinal cake-separated moxibustion,and the danazol group was treated with danazol suspension by intragastric administration,with two treatment courses. After treatment,the histomorphological changes of ectopic endometrium were observed by HE staining. The serum level of E2 was detected by ELISA,and the mRNA expressions of aromatase(P450arom)and cyclooxygenase-2(COX-2) in endometrium were determined by RT-PCR .   Results: The HE staining results showed that there was columnar hyperplasia in the epithelial cells of ectopic endometrium in the model group,with dense interstitial cells,local fibrosis and abundant blood supply. While the epithelial cells of ectopic endometrium were atrophic and presented the short cuboidal shape in the medicinal cake-separated moxibustion group,and the interstitial cells atrophy,reduced blood supply and local fibrosis were also observed. Compared with the blank control group,the level of serum E2 and the mRNA expressions of P450arom and COX-2 in ectopic endometrium were significantly increased in the model group(P<0.05). Compared with the model group,the level of serum E2 and the mRNA expressions of P450arom and COX-2 in ectopic endometrium were obviously decreased in the medicinal cake-separated moxibustion group and danazol group(P<0.05). Compared with the medicinal cake-separated moxibustion group,the level of serum E2 and the mRNA expressions of P450arom and COX-2 in ectopic endometrium were obviously decreased in the danazol group(P<0.05).   Conclusion: The medicinal cake-separated moxibustion may improve the blood circulation and decrease the expression levels of E2 in serum and P450arom and COX-2 in ectopic endometrium of rats with endometriosis,and thus inhibit the estrogen production and metabolism and suppress the growth of ectopic lesions.
