摘    要:目的:探索针刺实验文献中穴位生物学效应的分子机制,为临床及实验研究提供方法参考。方法:检索《中国知网》(CNKI)、《万方数据资源库》和《中国生物医学文献数据库》(CBM)三大数据库中的针刺动物实验文献。筛选出单纯针刺对动物机体内相关基因表达影响研究的文献,按文献纳入标准和排除标准选择、整理文献。采集文献中的有效穴位及相应的基因名称,运用Python编程语言编写的软件、Cytoscape3.3.0软件及MECODE复杂网络算法进行统计分析。结果:获得符合研究的文献2136篇,穴位233个,分子793个;频次排在前十的穴位为足三里、百会、内关、水沟、大椎、三阴交、肾俞、曲池、风府、阳陵泉;频次排在前十的分子有:BCL2、FOS、BDNF、Bax、CASP3、TNFA、GFAP、NGF、HSP70、IL1B;频次排在前五的穴位配伍有大椎-百会、水沟-百会、百会-足三里、三阴交-足三里、内关-水沟;穴位足三里处于穴位-基因网络的中心位置;通过复杂网络的模块分析发现,一些穴位与一些通路相关的基因落入同一网络模块中。结论:本研究发现足三里穴是针刺实验研究的最常用穴位,足阳明胃经可能与代谢通路关系密切,这一发现可为临床及实验研究提供新的研究思路。

Abstract:This paper was aimed to study the molecular mechanism of acupoint biology effect on acupuncture experimentarticles. The acupuncture experiment articles in CNKI, Wanfang Data Resource Library and the China BiomedicalLiterature Database (CBM) were searched. The literature on acupoint and related genes were selected according to theliterature inclusion criteria and exclusion criteria. The acupoint and corresponding genes in the literature were collectedand analyzed by software written in Python, Cytoscape 3.3.0 and MECODE algorithm. The results showed that 2136articles were collected, with 233 acupoints and 793 genes. Acupoints in the top 10 frequency were ST36 (Zusanli), GV20(Baihui), PC6 (Neiguan), GV26 (Shuigou), GV14 (Dazhui), SP6 (Sanyinjiao), BL23 (Shenshu), LI11 (Quchi), GV16(Fengfu) and GB34 (Yanglingquan). Genes in the top 10 frequency were BCL2, FOS, BDNF, Bax, CASP3, TNFA, GFAP,NGF, HSP70 and IL1B. Acupoint-groups in the top 5 frequency were GV14 (Dazhui)-GV20 (Baihui), GV26 (Shuigou)-GV20 (Baihui), GV20 (Baihui)-ST36 (Zusanli), SP6 (Sanyinjiao)-ST36 (Zusanli), PC6 (Neiguan)-GV26 (Shuigou). ST36(Zusanli) was in the center of the acupoint-gene network. Through module analyzing, there were some genes belong todifferent pathways and some acupoints in one network module. It was concluded that ST36 (Zusanli) was the coreacupoint in the acupoint experiment study, the stomach meridian of foot Yangming may be closely related with themetabolic pathway. This finding may provide new research ideas for clinical and experimental research.
