电针对慢性应激抑郁模型大鼠海马cAMP、cGMP含量的影响

摘    要:目的通过电针刺激印堂、百会穴对慢性应激抑郁模型大鼠第二信使环磷酸腺苷(cAMP)、环磷酸鸟苷(cGMP)含量影响的研究,揭示电针治疗抑郁症的可能作用机制。方法雄性SD大鼠32只,随机分为4组,每组8只,分别为正常对照组、模型组、电针组、氟西汀组。除正常对照组外,其余各组大鼠进行孤养,并接受慢性不可预知性应激刺激方式造模。各组大鼠分别于应激前1天、应激第21天后进行开野试验,观察大鼠的行为学变化。运用放射免疫法检测大鼠海马cAMP、cGMP含量。结果与正常对照组比较,模型组水平运动及垂直运动均显著减少(P0.01),大鼠探索能力降低;与模型组比较,电针组与氟西汀组水平运动和垂直运动均明显升高(P0.05),大鼠活动度增加。与正常对照组比较,模型组大鼠cAMP含量降低,cGMP明显升高,cAMP/cGMP比值减小,差异均有统计学意义(P0.01);与模型组比较,电针组和氟西汀组cAMP含量增加,cGMP含量降低,cAMP/cGMP比值增大,差异均有统计学意义(P0.05)。结论电针治疗抑郁症的可能作用机制之一是调节中枢cAMP、cGMP含量趋于平衡状态。

Abstract:  Objective:To study the effects of electroacupuncture on Yintang(EX-HN3)and Baihui(GV20)acupoints on the contents of second messengers cyclic adenosine monophosphate(cAMP)and cyclic guanosine monophosphate(cGMP)in model rat of chronic stress depression,and reveal the possible mechanism of electroacupuncture in the treatment of depression. Methods32 Sprague-Dawley male rats were randomly divided into the normal control group,model group,electroacupuncture group and fluoxetine group,8 rats in each group. Except the normal control group,the rats in the other groups were raised separately and treated with the chronic unpredictable stress stimulation to establish the model. One day before and 21st day of after stress stimulation,the rats in each group were performed the open field test respectively,and the behavioral changes of rats were observed. The contents of cAMP and cGMP in hippocampus of rats were detected by radioimmunoassay.  Results:Compared with the normal control group,the horizontal and vertical movement of rats in the model group were significantly decreased(P<0.01),and the exploration ability of rats was decreased. Compared with the model group,the horizontal and vertical movement of rats in the electroacupuncture group and fluoxetine group were obviously increased(P<0.05),and the activity of rats was increased. Compared with the normal control group,the cAMP content was decreased and the cGMP content was increased in the model group,and the ratio of cAMP/cGMP was reduced,with statistically significant differences(P<0.01). Compared with the model group,the cAMP content was increased and the cGMP content was decreased in the electroacupuncture group and fluoxetine group,and the ratio of cAMP/cGMP was increased,with statistically significant differences(P<0.05).  Conclusion:Regulating the contents of cAMP and cGMP in central nervous system to balanced state is one of the possible mechanisms of electroacupuncture in the treatment of depression.
