黄芪失笑散对急性心肌梗死大鼠Fas、FasL表达的影响

摘    要:目的:观察不同剂量黄芪失笑散对急性心肌梗死大鼠梗死边缘区细胞凋亡及Fas、FasL蛋白表达的影响.方法:取雄性SD大鼠60只,随机分为黄芪失笑散高剂量组、黄芪失笑散中剂量组、黄芪失笑散低剂量组、模型组和对照组.连续灌胃7d后,结扎冠状动脉左前降支造成急性心肌梗死模型,对照组不结扎.手术后4.5h留取心脏标本,采用原位末端标记染色法(TUNEL)检测细胞凋亡指数,采用Western blot法检测Fas、FasL蛋白表达量.结果:与对照组比较,黄芪失笑散各组可明显降低心肌细胞凋亡指数和Fas、FasL的蛋白表达量(P<0.01).不同剂量组间差异无统计学意义(P>0.05),且黄芪失笑散大剂量组效果最好.结论:黄芪失笑散能够保护急性心肌梗死大鼠的缺血心肌,其作用机制可能与降低梗死边缘区Fas、FasL蛋白表达水平从而抑制心肌细胞凋亡有关.

Abstract:Objective: To observe the effect of different doses of Huangqi Shixiao Powerder on the expression of Fas and FasL of Rats with acute myocardial infarction.Methods: 60 male SD rats were randomly divided into high dose group of Huangqi Shixiao Powerder,medium dose group of Huangqi Shixiao Powerder,low dose group of Huangqi Shixiao Powerder,model group and control group.7 d after gastric perfusion,coronary artery ligation of the left anterior descending branch was taken to make acute myocardial infarction model,the control group was not given ligation.4.5 h after the surgery,the heart specimens were kept and TUNEL were used to detect the cell apoptosis index,Western blot method was used to test the protein expression of Fas,FasL.Results: Compared with the control group,Huangqi Shixiao Powerder can obviously reduce the myocardial cell apoptosis index and the protein expression amount of Fas,FasL(P<0.01).The difference between different dose groups was not statistically significant(P>0.05),and the high dose group of Huangqi Shixiao Powerder had the best effect.Conclusion: The high dose group of Huangqi Shixiao Powerder can protect the ischemic myocardium of rats with acute myocardial infarction,the mechanism may be related with its function of reducing the protein expression level of Fas,FasL in the infarction edge area,thus inhibiting the myocardial cell apoptosis.
