丹参酮对大鼠实验性脑梗塞的防治作用

摘    要:凝闭大鼠一侧大脑中动脉、造成局灶性脑梗塞后24小时,大鼠外周中性粒细胞活性增强;缺血区脑组织脂质过氧化损伤严重;TTC染色后可见边界明显的脑坏死区。丹参酮可抑制脑缺血所致的中性粒细胞吞噬化学发光增强,减少白细胞在缺血区内的浸润,减轻脑组织脂质过氧化损伤,保护SOD活性,缩小梗塞范围。其抑制粒细胞功能激活与减轻脑缺血性损伤之间有显著的正相关关系。这一作用与地塞米松相似。提示:丹参酮抑制白细胞功能的作用可能是其防治脑梗塞的机制之一。

Abstract:Twenty four hours after occluding of the middle cerebral artery in rats, the circulating neutrophils were in activated state and the lipidperoxidation increased in ische- mic brain tissue. Tanshinone inhibited the chemoluminescence of the circulating neutrophil phagocytosis and decreased leukocyte infiltration in infarcted brain tissue. It also decreased lipoperoxide, malondialdehyde and increased SOD activity in ischemic cerebral region. In addition, it reduced cerebral infarction size in the model. The correlation between inhibitory effects on leukocyte and reduction in ischemic cerebral damage was significant. Dexamethasone used as a positive control had similar effects on neutrophils and ischemic cerebral injury. It was shown that the prophylactic effcts of Tanshinone on cerebral infarction might be mainly due to its inhibition on leukocyte function.
