摘    要:为改进HPLC测定氟尿嘧啶 (5 fluorouracil,5 FU)的方法 ,选择对氨基苯甲酸 (PABA)为内标物 ,用C18色谱柱 ,流动相甲醇 磷酸缓冲液 (pH 6.8) ,检测波长 2 70nm ,测定血清中 5 FU含量。结果 5 FU的最小检出限量为 1 .2ng,5 FU及PABA的萃取回收率分别大于 88.0 1 %和 86.5 5 %。

Abstract:To improve the determination of 5-FU by high-performance liquid chromatography, PABA was selected as an internal standard in the analytical method. For determination of 5-FU concentration in serum, a reversed-phase C 18 column with a mobile phase consisting methanol-phosphate buffer (pH 6.8) was used. UV detector was set at 270 nm. The limit of determination for 5-FU was 1.2 ng. The extraction recovery of 5-FU and PABA were more than 88.01% and 86.55% respectively.
