紫杉醇防治移植动脉硬化作用的实验研究

摘    要:目的采用大鼠胸腹主动脉移植模型,探讨紫杉醇防治移植动脉硬化的作用。方法Wistar大鼠40只,SD大鼠16只,按供、受鼠不同分为4组:A组为假手术组;B组为同品系移植(Wistar to Wistar)组;C组为单纯异品系移植(Wistar to SD)组;D组为异品系移植(Wistar to SD)紫杉醇干预组。移植术后30天切取移植动脉,采用光镜和电镜定性观察,并用计算机图像分析系统定量观测紫杉醇对移植动脉的影响。结果与A组比较,术后30天B组动脉壁无增厚、血管内膜未见明显增生;C组内膜增生明显,内外膜有大量炎症细胞浸润。紫杉醇则显著抑制移植动脉内膜增生,并减轻移植动脉内外膜炎症细胞浸润,防止管腔狭窄。结论紫杉醇可明显抑制异品系大鼠移植动脉内膜增生和炎症细胞浸润,防治移植动脉硬化。

Abstract:Purpose To study the effect of antiproliferation agent paclitaxel on preventing graft arterioscleriosis in rat aortic transplantation model. Methods Forty inbred Wistar and 16 SD rats were randomly divided into 4 groups. A was the sham group; B was the control group of isografts ; C was the allografts control group ; D was the allografts treated group with paclitaxel. All recipients were killed 30 days after transplantation. The specimens were studied by light and electronic microscopy. Regional changes in the lumen,intimal and medial layers were measured with computer imaging analysis system. Results Thirty days after transplantation,little intimal thickening and inflammatory cells infiltration in neointima and adventitia were showed in group B as well as group A. But pronounced intimal thickening and inflammatory cells infiltration in neointima and adventitia were seen in group C compared with group B. Intimal thickening was significantly reduced and inflammatory cells were suppressed in allogeneic grafts after treated with paclitaxel. Conclusions Paclitaxel can obviously suppress the hyperplasia of intima and inflammatory cells infiltration, and prevent the graft arteriosclerosis.
