温郁金二萜类化合物C对人胃癌SGC-7901细胞中Caspase-9,3,7和PARP(89KD)蛋白表达的影响

摘    要:目的:探讨温郁金二萜类化合物C1]对人胃癌SGC-7901细胞中Caspase-9,Caspase-3,Caspase-7和PARP(89KD)蛋白表达的影响。方法:用从温郁金醚提物中提取得到的二萜类化合物C分别以0、40、55、70μg/mL 4个浓度作用于人胃癌SGC-7901细胞24h;用Western Blot杂交法检测4个浓度组中人胃癌SGC-7901细胞的Caspase-9、Caspase-3、Caspase-7和PARP(89KD)蛋白量的表达。结果:温郁金二萜类化合物C能上调SGC-7901细胞中Caspase-9、Caspase-3、Caspase-7及PARP(89KD)蛋白的表达,与空白对照组相比,差异具有统计学意义(P<0.05);且具有一定的剂量依赖性。结论:温郁金二萜类化合物C可通过上调Caspase-9,Caspase-3,Caspase-7,PARP(89KD)的表达来诱导人胃癌SGC-7901细胞发生凋亡。

Abstract:Objective: To investigate the effect on expression of Caspase-9,3,7,and PARP(89KD) of SGC-7901 by diterpenoid curcumrinol C from ether extract of Radix Curcumae.Methods: SGC-7901 cells were treated by curcumrinol C on four different concentrations(0,40,55,70μg/mL) and Western Blot hybridization was used for detecting expression of Caspase-9,3,7 and PARP(89KD) of SGC-7901 cells after treated by curcumrinol C.Results: Compared with the control group,curcumrinol C could increased the expression of Caspase-9,Caspase-3,Caspase-7 and PARP(89KD)(P<0.05),which showed a dose-dependent.Conclusion: Curcumrinol C can induce tumor cells apoptosis by increasing the expression of Caspase-9,Caspase-3,Caspase-7 and PARP(89KD).
