食管鳞状细胞癌中AEG-1的表达和微血管密度与肿瘤转移及预后的关系

摘    要:目的探讨食管鳞状细胞癌(ESCC)组织中星形细胞上调基因-1(AEG-1)的表达与其生物学行为以及微血管密度(MVD)之间的关系,分析AEG-1的表达及新生血管形成在食管癌侵袭、转移及预后中的生物学意义。方法应用免疫组织化学方法(SP法)检测163例ESCC组织、20例非癌食管组织中的AEG-1及CD105标记的MVD值,并分析AEG-1的表达及MVD与组织学分级、淋巴结转移的相关性。结果 AEG-1在ESCC组及淋巴结转移组的阳性表达明显高于非癌组及无淋巴结转移组,差别有统计学意义(P<0.05),但组织学分级各组间差别无统计学意义(P>0.05)。CD105标记的MVD在ESCC组明显高于非癌组,在淋巴结转移组明显高于无淋巴结转移组,差别有统计学意义(P<0.05);组织学分级各组间差别无统计学意义(P>0.05)。AEG-1表达与MVD呈正相关(r=0.428,P<0.05)。结论 AEG-1高表达可能与肿瘤血管发生及肿瘤转移密切相关,可作为判断食管癌患者预后的参考指标。

Abstract:Objective To investigate the correlation of expression of astrocyte-elevated gene-1(AEG-1) and microvessel density(MVD) with clinical biological characteristics,and to analyze the relationship of these indicators with the invasion,metastasis and prognosis of esophageal squamous cell carcinoma.Methods High sensitive SP immunohistochemical method was used to detect the expression of AEG-1 and CD105 in 163 cases of ESCC and 20 cases of para-cancerous normal esophagus tissues.The relationship between the expression of these indicators and differentiation and lymph node metastasis was investigated.Results The positive rates of AEG-1 expression in cases with lymph nodal meatstasis was dramatically higher than that in the normal tissues and without lymph nodal metastasis(P<0.05),but was not correlated with the histology grade(P>0.05).The MVD in ESCC was dramatically higher than that in the normal tissues and without lymph nodal metastasis(P<0.05),but was not correlated with the histology grade(P>0.05).AEG-1 expression was closely correlated with MVD(r=0.428,P<0.05) Conclusions AEG-1 expression may be related to tumor angiogenesis and progression and is a valuable prognostic factor in patients with ESCC.
